Quantum dots (QDs) are currently one the most widely used fluorescent nanoparticles employed for labelling as well as imaging purposes. The preparation of QDs is commonly done by organic or aqueous synthesis. Moreover, biosynthesis of QDs by numerous organisms (bacteria, yeasts, earthworms and/or plants) has been described. The synthesis by UV light illumination possesses variety of advantages including simplicity and short time of preparation. In this work, a systematic study of preparation of aqueous CdTe, CdSe, ZnS and ZnSe QDs capped by mercaptosuccinic acid is presented. The nanoparticles were prepared by UV light (254 nm and 312 nm) illumination and characterized by UV/vis spectrometry, fluorescence spectrometry, particle size distribution and capillary electrophoresis with light emitting diodeinduced fluorescence detection (λex=400 nm). It was found out that the maximum fluorescence intensity was achieved in case of CdTe after 30 minutes of illumination and the emission maximum of obtained QDs was 510 nm. These QDs exhibited also the narrowest emission spectra (FWHM = 40 nm). In contrary, the ZnS and ZnSe QDs reached the maximum of fluorescence intensity already in 10 minutes of illumination; however, the FWHM was 130 – 135 nm and the emission maximum was in the range from 430 to 450 nm. It can be concluded that UV illumination is an alternative method of synthesis of QDs with the advantage of production of QDs with consistent emission maximum
