Original Abstract

Biotechnological production of PHA employing extremophiles within concept of Next-Generation Industrial Biotechnology (NGIB) bring many benefits to whole process considering requirement for sterility, cultivation, and also specific possibilities for PHA isolation process. Within our work, we focused on two extremophilic producers, halophilic bacterium Halomonas halophila (CCM 3662) and thermophilic bacterial strain Schlegelella thermodepolymerans (DSM 15344). Unique features of selected extremophiles are used for development of environmentally friendly and also economically feasible isolation approach combining hypotonic shock, elevated temperature and detergent sodium dodecyl sulphate (SDS), which can lead to PHB with purity higher than 90 wt.%. Moreover, we will focuse on recyclation of residual SDS, which represents pollutant of wastewater after isolation.
