In this work, we performed a sequential chemical extraction with a series of organic solvents with an increasing polarity (trichloromethane – TCM1 < ethyl acetate – EAC2 < acetone – ACE3 < acetonitrile – ACN4 < n-propanol – PRO5 < methanol – MET6) on Czech biochar and subjected the obtained organic fractions to a physicochemical characterization utilizing thermal and spectrometric methods of structural and compositional analysis. Spectrometric techniques such as Ultraviolet-visible (UV/Vis) and Attenuated Total Reflectance (ATR-FTIR) were combined with thermogravimetric and elemental analysis of the organic fractions. Biochar was homogenized and 5000 mg was then inserted in a thimble, and Soxhlet sequential extracted for 24–168 h. The organic fractions isolated from the biochar were concentrated using a rotary evaporator. Furthermore, the brown to black extracts were dried in a vacuum desiccator filled with anhydrous sodium sulphate as a drying medium. In total, the fractionation procedure extracted only 6.8 (wt. %) of the initial Czech biochar. The extraction yields of organic fractions ranged from 0.3–3.6% in the following order samples: (MET6 >> TCM1 > EAC2 ~ ACN4 > ACE3 > PRO5). Obvious differences in chemical properties of individual organic fractions were found. While the strong polar solvent used in the last step preferentially released organic compounds rich in carboxylate functional groups and ash moieties, apolar solvents (TCM1–EAC2) provided lipid-like aliphatic components, which are predominantly substituted with ester and ether functional groups. The fraction ACE3 shows structural features with its higher aromaticity and high content of amide functional groups such as protein-like structural moieties.
