Original Abstract
DNA methylation plays a crucial role in epigenetic modification which regulates gene expression and genome stability. Application of Pacific Biosciences sequencing technology (PacBio) and Oxford Nanopore sequencing technology (ONT) has shown promising approaches for detecting single base modification for DNA methylation. In this article, we analysed the base modification detection on ONT sequences and then compared it with PacBio analysed data. We found that both Nanopore and PacBio methods were effective but showed variations in their result. We analyzed the methylation pattern in Clostridium beijerinckii NRRL B-598 and found that the ONT data in combination with tombo tool can help detect modification at low coverage sites while avoiding false reading at these areas. Our study provide insight into these two approaches of DNA methylation detection, which will help in guiding the choices of appropriate method for specific research purposes.
