Original Abstract
Ceramic-polymer nanofibers were prepared by the electrospinning method with the aim to prepare a biocompatible and bioactive structure usable as scaffold. Hydroxyapatite nanoparticles were chosen as the ceramic fillers of the polymer fibers composed of the polycaprolactone. The diameter of the prepared fibers was approximately 180 nm. The hydroxyapatite nanoparticles had needle-like shape. The needles were approximately 30 nm in the diameter and 300 nm long. Thanks to theirshape, they were easy to be incorporated into structure of the fibers. Dispersion of the particles in the solution was improved by plasma and ultrasound treatment ofthe precursor. Cytotoxicity tests were performed on the prepared fibers. Specific growth rate (proliferation rate of the cells) was evaluated on the samples. The results were compared with the results of baseline samples (slide glass) and polycaprolactone samples. It was found out that presence of the ceramic compound supports activity of the cells. Another improvement of the specific growth rate was reached by low-temperature plasma treatment of the fibers. Since standardized cytotoxicity testing is executed on the thinlayer of the material which is transparent for visible light, it was important to test “bulk” structure. The bulk structure simulates future utilization of the material as scaffolds. Therefore, thick layer of plasma-treated composite structure and polycaprolactone multi-layered fibrous structures as well as untreated polycaprolactone structure were tested. It was found out that that plasma treatment and ceramic particles in the structure have a great impact on the proliferation rate of the cells.
