Original Abstract
Polyhydroxyalkanoates (PHA) are microbial polyesters produced by various bacteria. PHA is good alternative to petrochemical polymers and is also totally biodegradable, biocompatible and renewable. Extremephilic organisms are organisms which live and reproduce under extreme conditions. Extreme conditions may be e.g. high/low temperature, high/low pH or higher salinity. This extreme cultivation conditions have positive influence for biotechnology price because cultivation can be operated in semi- or even non-sterile conditions. We focused on isolation bacteria from compost and activated. We used our original isolation protocol for isolation of thermophilic bacteria accumulating PHA. Isolation protocol used application of changing of osmotic pressure because PHA granules in bacteria protect them from fluctuations in osmotic pressure. Furthermore, we focused on thermophilic isolation for thermophilic producers. One of the isolates accumulated PHA up to 60 % of cell dry weight. Further, when we used proper precursors, it was also capable produced copolymer with 3-hydroxyvalareate or 4-hydroxybutyrate units but also produced terpolymer. These copolymers and terpolymer reveal very interesting mechanical and thermal properties
