Postoperative complications in bone tissue healing often include inflammatory bacterial infections like osteomyelitis, commonly caused by gram-positive bacteria such as Staphylococcus aureus (SA). Antibiotics applied as part of so-called bone cements are commonly used to treat these infections. However, the current market lacks a bone cement capable of gradual antibiotic release at the infection site due to its material composition. This work focused on the evaluating the impact of various factors on the rate and release method of the selected antibiotic vancomycin incorporated into a newly developed resorbable bone cement based on ceramics and a thermosensitive copolymer. First, the influence of the method of preparation and mixing of bone cement on the internal structure, morphology, and pore size was monitored using two imaging techniques, namely scanning electron microscopy and micro-CT. Hand mixing resulted in finer pores (30–250 µm), while electric mixing shifted the pore size distribution to a range of 50–400 µm. Subsequently, the antibiotic release kinetics of the prepared bone cements were measured under physiological conditions at 37°C utilized UV-VIS spectroscopy and liquid chromatography. The study revealed that the preparation method of bone cements did not significantly affect the kinetics of antibiotic release, as well as measurements under static or dynamic conditions. However, antibiotic concentration and the amount of bone cement used had a substantial impact on antibiotic release. It was found that the release pattern followed first-order kinetics, with 50% released at the lowest antibiotic concentration and up to 90% at the optimal 1% concentration. The newly developed resorbable bone cement demonstrates promise as a material for controlled antibiotic release, particularly in treating inflammatory bone infection in medical specialties like traumatology or orthopaedics.
