Basics of Analytical Chemistry

The aim of the course is to give students an basic overview of the structure and content of classical analytical chemistry and of the basic methods and techniques of instrumental analytical chemistry.
The course consists of the following thematic units:
Basic notions in analytical chemistry. Explanation and analytical aspects of chemical equilibrium. Classification of methods of classical qualitative and quantitative analysis. Principles of selected instrumental methods. Electroanalytical methods. Optical methods, their classification, properties of electromagnetic radiation. Principles and importance of chromatographic and electromigration methods. Introduction to analytical chemometry. Evaluation of analytical results. Analytical errors and methods of quantitative evaluation.

Good knowledge of common and inorganic chemistry.

Rules for evaluation and completion of the course

Course credit is awarded for 100% attendance at seminars (or properly excused absence).
Credit may be obtained for successful completing of partial written tests during the semester (ie getting at least 6 points out of 10 possible). In case of failure in one partial test (or more) course credit could be awarded for the successful completing of the final test (ie, obtaining at least 60 points out of 100).

Examination consists of a written and oral part.

Attendance at lectures is recommended, but not controlled.
Attendance at seminars is compulsory and is checked.
The students' work is controlled by the partial written tests during the semester in computational exercises.
Consultations are provided on request.

Aims

The aim of the course is to give students overview about analytical chemistry, classical analysis, selected instrumental analytical methods from the field of electrochemistry, spectrophotometry and separation techniques and chemometric approaches for processing the results.
Increased knowledge, skills and competencies of students will be reflected mainly in the following areas:

1.Students will became acquainted with the theoretical foundations of analytical chemistry.
2. Students will have cognizance of classical qualitative and quantitative analysis methods.
3. They will know the principles of simple instrumental analytical methods and possibilities of their application.
4. They will adopt the basic principles of chemometrictreatment of analytical results obtained using simple instrumental-analytical methods.
5. Students will improve their skills in computing solutions to solve simple analytical problems.

Syllabus

1.-2. Roztoky. Hmotnostní zlomky, látková množství, koncentrace, obsahy analytu ve směsích, směšovací rovnice. Iontová síla roztoků, aktivitní koeficienty, aktivitní a koncentrační rovnovážné konstanty.

3. Volumetrická a gravimetrická stechiometrie.

4.-5. Acidobazické reakce. Výpočty pH silných a slabých elektrolytů, kyselin, bazí, solí, amfolytů. Acidobazické tlumiče, jejich pH a kapacita. (Distribuční diagramy)

6. Komplexotvorné reakce. Výpočty koncentrací analytů v roztocích komplexů, distribuční koeficienty komplexů ve směsích, konstanty stability komplexů.

7. Reakce za vzniku sraženin. Součin rozpustnosti a rozpustnost iontových sraženin. Vlivy dalších iontů na rozpustnost.

8.-9. Redoxní reakce. Oxidačně-redukční potenciály, konstanta redoxních rovnováh. Výpočty z průběhu reakce dvou redoxních párů. Výpočty koncentrací analytů z rovnic pro Nernstovy potenciály.

10.-12. Základní výpočty z oblasti elektroanalytických, spektrofotometrických a separačních metod.

13. Příklady základního statistického vyhodnocování analytických výsledků.
