The problem of pure DNA preparation from complex matrix and the effects of the inhibitors from complex biological samples can be reduced by the selection of the appropriate nucleic acid isolation method. Numerous solid phase systems, especially magnetic particles, have been used to a great extent for DNA isolation. Magnetically driven separation techniques using micro- and nano- sized magnetic solid carriers have become one of modern methods to speed up and facilitate the previously used separation and purification procedures. Magnetic particles are mostly based on magnetic iron oxide (magnetic core) covered by polymer surfaces suitable for modification by different functional groups (e.g. –COOH, -NH2). Free or incomplete encapsulated cores and some components applied in the preparation of magnetic particles can interfere with polymerase chain reaction (PCR). Magnetic particles with complete covering of magnetic nuclei do not interfere in PCR, and thus such particles are preferred.
