Sponges are increasingly recognized as key ecosystem engineers in many aquatic habitats, playing important roles in ecological processes. As filter-feeders, they capture and transform a suite of organic and inorganic nutrients in their tissue1. To understand the water flow through the sponge, it is necessary to quantify and visualize the multiscale 3D structural morphology of the sponge’s internal organization, and characterizing the complex canal system within the sponge’s tissue. It is also important to identify and image the 3D organization of the individual choanocyte and the chambers they form. as this has influence on the required pressure gradient for the water flow within the whole sponge. Given the complex structure of Sponge’s and phenomenological similarities to other organic and/or bio-synthetic materials systems2 , a full 3D correlative characterization, where precise and reliable identification and tracking of multiple volume-based regions of interest throughout scales and imaging modalities will be beneficial and necessary. The use of newly developed CoSMic module ( 3D Correlative Spectroscopy Microscopy Toolkit) for AVIZO/AutoScript platforms will allow such a 3D correlative approach as shown in figures 1 and 2. As shown in figure 1, two Areas of Interest (AOI) are defined by the user within the μCT volume and are automatically targeted and patterned within Hydra, using the CoSMic Module . The required surrounding areas to AOI1 is then digged and prepared automatically via CoSMic, in order for further ASV jobs to be setup and executed by the user on Hydra.
