Skin wound healing is a complex process involving the interaction of cells, mediators, cytokines, matrix remodeling, and the neurovascular system. The success rate of skin healing has significantly increased with the use of custom-made skin substitutes that mimic the extracellular matrix (ECM) and its properties. Collagen, the most abundant protein in the human body, serves as a structural building block of the ECM in most native tissues. It possesses unique structural properties essential for skin tissue engineering, regulating functional responses while being low-inflammatory, biodegradable, biocompatible, and bioresorbable. Chitosan and oxidized cellulose also exhibit various beneficial biological properties, such as biocompatibility, good interaction with cells, low immunogenicity, and antimicrobial activity. Additionally, pro-healing growth factors and antibacterial catechol additives are known to enhance the biological activity of these materials. In this research, we enriched cross-linked, lyophilized 3D porous collagen-polysaccharide carriers with both pro-healing proteins and antibacterial additives top-coated with bioresorbable nanofiber layer to evaluate their mechanical stability, strength, absorbability, in vitro cytotoxicity, angiogenic, and antibacterial potential. The prepared bilayer scaffold demonstrated ideal hydrolytic and enzymatic stability in vitro, was easy to handle without signs of deformation, and exhibited both antimicrobial and biocompatibility properties necessary for wound healing management.
