Vascular endothelium play important role in many physiological and pathological states and it is also pivotal structure allowing efective drug tansfer invivo – both in form of soluble macromolecules or in form of encapsulated molecules (liposomes or another nature or synthetic nanovesicles). Endothelial cells (ECs) form layer with specific permeability, which react to different biophysical and biochemical state during. Fluorescent dyes and spectrometer traditionaly help to quantify leakage kinetics across the the model layer of ECs in vitro under normal or some pathological conditions (infection, hypoxic, acidic condition). We have focused to construct inovative two-comparmtents in vitro model and to test leakage kinetics of modern liposomes with miRNA cargo and to measure accelerating effect of ultrasound stimuls on ECs.
