Laboratory Classes in Organic Chemistry

Practical training at isolation, separation and identification procedures used in organic laboratories within the frame of representative one-step or multi-step syntheses. Training of the basic lab techniques of the synthesis and the isolation of organic compounds.

Learning outcomes of the course unit

This course provides students especially the ability to:

Working with laboratory equipment and measuring instruments for conventional organic syntheses.
Experience with preaparation of organic substances in the laboratory scale.
Experience with occupational safety and health in the chemical laboratory work with organic compounds.

Planned learning activities and teaching methods

The course uses teaching methods in form of Laboratory exercise - 4 teaching hours per week. The e-learning system (LMS Moodle) is available to teachers and students.

Assesment methods and criteria linked to learning outcomes

To obtain the credit, the student must:
Passing 11 jobs during the semester.
To master the theoretical principles of tasks.
Submit prepared and adjusted products.
Submit a fully developed laboratory protocols.;

Course curriculum

Students are briefed by organization of laboratory and syntheses in detail during the first lab. Safety training is also realized. Students realize at least 11 syntheses of the following:
1. beta-Nitrostyrene and 1,5-diphenylpenta-1,4-dienone (nucleophilic aditions on carbonyl compounds)
2. meta-Dinitrobenzene (elektrophilic aromatic substitution, nitration to the second step)
3. Di-n-butylether (preparation of ether by dehydratation of alcohol)
4. Ethylbromide and terc-butylchloride (SN2 and SN1 reaction)
5. Anhydride of bicyklo[2.2.1]hept-2-ene-5,6-dikarboxylic acid and bicyklo[2.2.1]hept-2-en-5,6-dikarboxylic acid (Diels-Alder reaction)
6. 4-Jodonitrobenzene and 1-[(4-nitrophenyl)azo]-2-naphtol (diazotation of aromatic amines, Sandmeyer reaction and copulation)
7. Ethyl-acetate (Fisher's esterification)
8. Benzylidenphenylamine and benzylphenylamine ( condensation of amine with carbonylic compound, reduction of double bond)
9. N-(4-Hydroxyfenyl)acetamid (Paracetamole) a N-(4-Ethoxyfenyl)acetamid (Phenacetine) (acylation of the amine and Wiliamson's synthesis of ether)
9. Benzimidazole a benzotriazole (preparation of simple heterocarbohydrates)
10. 3-Nitroaniline (partitial reduction of nitrocompound)
11. Diphenylmethanol (benzhydrole) (preparation and reaction of Grignard's reagents)
12. 2-(4-Methylbenzoyl)benzoic acid (Friedel-Crafts acylation)

Labs at last week of the term are substitute for unqualified or sick students during the term.

Aims

The basis of the course is to enhance the adoption and expansion of basic laboratory methods and techniques (eg work at reduced temperatures, vacuum distillation, steam distillation) in the synthesis and analysis of organic compounds (determination of melting point, refractive index). Tasks are chosen so as to include the most common and most important types of chemical reactions discussed in hours of organic chemistry.
