Laboratory Classes in Microbiology

The basic rules of safety work in microbiological laboratory. Preparation of culture media, various sterilization methods. Inoculation, incubation, isolation of microorganisms. Preparation of smears for staining, microscopic observation. The Gram´s staining method, several other special staining techniques. Quantification techniques - direct and indirect method. Yeasts. Fungi. The microbiological analysis of water and soil. The microbiological control of foods.

Learning outcomes of the course unit

Practical knowledge and skills of basic microbiological laboratory methods: preparation of culture medium, inoculation, incubation and isolation of microorganisms, preparation of smears for staining, various staining methods, microscopic observation of microorganisms.
The ability to apply these skills for the microbiological examination of foods, water and other materials.

Prerequisites

Successful completion of practical classes is conditional on the basic theoretic knowledge of general microbiology.

Planned learning activities and teaching methods

The course uses teaching methods in form of Laboratory exercise - 3 teaching hours per week. The e-learning system (LMS Moodle) is available to teachers and students.

Assesment methods and criteria linked to learning outcomes

Conditions for awarding the course unit credit: 100 % participation in laboratory classes, elaboration and submitting of reports in written form and credit test (at minimum 70 %).

Course curriculum

The basic rules of safety work in microbiological laboratory.
1. Preparation of sterile glassware and other tools
2. Preparation of culture media and its sterilization
3. Isolation, inoculation and storage of microorganisms. Cultivation conditions
4. Microscopic observation of microorganisms.
5. The negative staining, staining of bacterial capsules and spores
6. Quantification of microorganisms by direct microscopic counts. Vital staining.
7. Quantification of microorganisms by colony count method.
8. Morphology of yeasts
9. The microbiological analysis of water
10. Morphology of fungi
11. The microbiological analysis of soil
12. The microbiological control of foods

The aim of these practical classes is to make students familiar with basic procedures, methods and techniques used in microbiological laboratory.

Attendance is required and checked by the teacher.
