Laboratory Classes in Organic Chemistry

Practical teaching of the preparation, isolation, separation and identification of organic compounds based on specific one-step or multi-step syntheses. Acquisition of basic and expanding laboratory techniques used in the synthesis and subsequent isolation of organic substances. The student also has the opportunity to familiarize himself with the basics of separation techniques (thin-layer and column chromatography) and the method of structural identification of organic molecules using NMR spectroscopy

Rules for evaluation and completion of the course

To obtain the credit, the student must:
Passing 11 jobs during the semester.
To master the theoretical principles of tasks.
Submit prepared and adjusted products.
Submit a fully developed laboratory protocols.;

Aims

The basis of the course is to enhance the adoption and expansion of basic laboratory methods and techniques (eg work at reduced temperatures, vacuum distillation, steam distillation) in the synthesis and analysis of organic compounds (determination of melting point, refractive index). Tasks are chosen so as to include the most common and most important types of chemical reactions discussed in hours of organic chemistry.
This course provides students especially the ability to:

Working with laboratory equipment and measuring instruments for conventional organic syntheses.
Experience with preaparation of organic substances in the laboratory scale.
Experience with occupational safety and health in the chemical laboratory work with organic compounds.

Syllabus

As part of the first internship, training on organization, familiarization with individual tasks and safety in the organic laboratory is carried out. In the following weeks, students will complete the tasks below:

1. Labor safety training in the laboratory 2. TLC basic technique for monitoring the progress of the reaction, stationary phase, solvent effect 3. NMR, basic sample preparation procedures, measurement procedure, familiarization with instrumentation 4. NMR, evaluation of measured data (FID), work from software 5. Ethyl bromide and tert-butyl chloride (SN2 and SN1 reactions) 6. Anhydride of bicyclo[2.2.1]hept-2-ene-5,6-dicarboxylic acid and bicyclo[2.2.1]hept-2-ene-5,6-dicarboxylic acid (Diels-Alder reaction) 7. 4-Iodonitrobenzene and 1-[(4-nitrophenyl)azo]-2-naphthol (diazotization of aromatic amines, Sandmeyer reaction and coupling) 8. Ethyl acetate (Fisher esterification) 9. Benzylidenephenylamine and benzylphenylamine (condensation of an amine with a carbonyl compound, reduction of a double bond) 10. N-(4-Hydroxyphenyl)acetamide (Paracetamol) and N-(4-Ethoxyphenyl)acetamide (Phenacetin) (Amine Acylation and Williamson Ether Synthesis) 11. Benzimidazole and benzotriazole (preparations of simple heterocycles) 12. 3-Nitroaniline (partial reduction of a nitro compound) 13. Diphenylmethanol (benzhydrol) (preparation and reaction of Grignard reagents) The last week of the semester is reserved for compensation for missed (due to illness) or unrecognized (e.g. insufficient knowledge, yields or non-compliance with safety rules) synthesis of the above-mentioned substances.z/elearning/course/view.php?id=
